绿色木霉内切纤维素酶的分离纯化及酶学性质的研究 被引量:3

摘要 绿色木霉AS3.5455经固体发酵,提取胞外纤维素酶,经硫酸铵分级沉淀、透析、DEAE Cellulose-52离子交换层析、SephadexG-150凝胶柱层析等纯化出单一组分的内切β-葡聚糖苷酶,经SDS-PAGE电泳分析,该内切酶的相对分子量约为58.7KD,内切β-葡聚糖酶的最适作用温度为55℃,最适反应pH为5.5。 Cellulase has been extracted from the solid media fermentated by trichoderma viride AS3.5455. Endo - β - glucanase has been isolated and purified by methods of ammonium sulfate precipitation, DEAE Cellulose - 52 chromatography and SephadexG-150 chromatography. The molecular weight of endo - 1, 4 - β - D - glucanase is 58.76KD determined by SDS - PAGE.The property of endo - 1, 4 - β - D - glucanase has been determined as well, and results indicate optimal reaction temperature 55 ℃, optimal pH5.5,
