摘要 目的:观察白介素-18(IL-18)对诱导体外培养的人脐动脉平滑肌细胞组织因子(TF)表达的作用。方法:在培养的人脐动脉平滑肌细胞中加入IL-18(浓度为100μg/L),用酶联免疫吸附分析法(ELISA)及细胞发色底物法分别测定培养基中TF抗原及细胞冻融液的TF活性。结果:培养基中TF抗原含量在4、6、8h组均高于24h组和对照组(P<0.05),且4h组最高(P<0.05),4h与6h2组间差别无统计学意义(P>0.05),8h组均低于4h组和6h组(P<0.05),24h组与对照组差别无统计学意义(P>0.05)。细胞冻融液中总TF活性在6、8、24h组均高于对照组(P<0.05),6h组和8h组均高于24h组(P<0.05),6h与8h组间差别无统计学意义(P>0.05)。结论:IL-18可以很快诱导体外培养的人脐动脉平滑肌细胞表达TF,使动脉血管平滑肌细胞呈现一过性促凝状态。 Objective: To investigate the effects of interleukin-18 (IL-18 )on the expression of tissue factor (TF)in the cultured umbilical arterial smooth muscle cells. Methods: The cultured human umbilical arterial smooth muscle cells were incubated with IL-18 (100μg/L). The tissue factor antigen in media was assayed by ELISA. The activity of rlT in cells lysates was estimated by chromogenic assay. Results: The antigen of TF in media was significantly higher in groups of 4 h, 6 h and 8 h than that in group of 24 h and controls (P〈 0.05),the highest level in the group of 4 h (P〈 0.05). There was no significant differences in antigen of TF between 4 h and 6 h groups (P 〉 0.05 )and between groups of 24 h and control (P 〉 0.05). The antigen of TF in media was significantly lower in group of 8 h than that in groups of 4 h and 6 h (P〈 0.05). The activity of TF in cell lysates was significantly higher in groups of 6 h,8 h and 24 h than that in controls (P 〈 0.05). The activity of TF was higher in groups of 6 h and 8 h compared with that in group of 24 h (P 〈 0.05). There was no significant difference between groups 6 h and 8 h(P〉 0.05). Conclusion" The tissue factor was rapidly induced in cultured umbilical arterial smooth muscle cells in vitro,which induce a transient procoagulant state on the surface of exposed smooth muscle cells.
