摘要
观察吡咯喹啉醌(pyrroloquinoline quinone,PQQ)对大鼠海马神经细胞结构的影响。侧脑室注射PQQ,60d后行脑组织切片并进行HE和尼氏染色,观察大鼠海马组织的形态学变化。用免疫组化法观察胆碱乙酰基转移酶的表达情况。结果表明,与对照组相比,PQQ处理后,大鼠海马神经细胞胞体和核增大,神经细胞的密度增加,尼氏小体的光密度值增加,此变化呈剂量依赖性。大剂量的PQQ处理后,海马神经元的胞体和核变小,细胞的凋亡率增加。胆碱乙酰基转移酶的表达量随PQQ刺激而增加。此结果表明,PQQ能促进鼠海马神经细胞的生长发育,但过大剂量PQQ则具有神经毒性作用。
To investigate the effects of pyrroloquinoline quinone (PQQ) on the morphusis of the hippocampal formation, PQQ was admin-istered into the rat lateral cerebroventricle. After 60 days, the hippocampal formation serially cut and observed by HE staining and Nissl' s staining, respectively. The expression changes of choline acetyhransferase (CHAT) was also detected by immunohistochemical staining. Compared with the control group, it was found that the size of the neurosoma and nucleus, the optical density of Nissl's body and the density of neurons were increased markedly after lower concentration of PQQ injection. However, after higher concentration of PQQ injection, the size of the neurosoma and nucleus changed into smaller and the apoptosis rate increased markedly. The expression of ChAT protein also increased dependent on injection of PQQ. These results indicate that PQQ can improve the growth and development of hippocampal neurons in vivo, but overdose PQQ can induce the neurotoxic effect.
