摘要
目的:探讨肿瘤坏死因子(TNF)、干扰素(IFN)和顺铂(DDP)、表阿霉素(EPI)联合应用对正常骨骼肌细胞的细胞毒作用,为骨恶性肿瘤的治疗提供理论基础。方法:用原代培养的Wister大鼠的骨骼肌细胞作为靶细胞,采用四甲基偶氮唑蓝试验法(MTT)观察TNF、IFN和DDP、EPI联合应用,在不同浓度和不同时间对正常骨骼肌细胞生长的影响。结果:TNF、DDP和EPI联合应用对正常骨骼肌细胞的生长起抑制作用,TNF、IFN、DDP和EPI联合应用,在浓度为250U/ml+0.0005g/L+0.0005g/L时可促进骨骼肌细胞的生长与增殖,从浓度为500U/ml+0.001g/L+0.001g/L时转为抑制骨骼肌细胞的生长。结论:TNF、IFN、DDP和EPI联合应用,在低浓度时对正常骨骼肌细胞的生长具有促进、增殖作用,高浓度时可抑制正常骨骼肌细胞的生长作用。
Objective: To explore the cytotoxic effect of tumor necrosis factor and interferon and chemotherapeutic drugs on normal skeletal muscle cells in order to give more information for treating bone tumor in the clinic. Methods: The wister rat's skeletal muscle cells were cultured as target cells. The MTT assay was used to detect the different effects on normal skeletal muscle cells when different concentration TNF and/or IFN and DDP and /or EPI were used in different times. Results: There was inhibitory effec,t on normal skeletal muscle cell's growth when TNF combined with DDP and EPI were used. There was positive effect on normal skeletal muscle cell's growth when TNF was combined with IFN and DDP, and EPI was used in low concentration (250 U/ml+0. 000 5 g/L+0. 000 5 g/L) and inhibitory effect in high concentration (500 U/ml+0. 001 g/L+0. 001 g/L). Conclusion: There is inhibitory effect on normal skeletal muscle cell's growth when TNF is combined with DDP and EPI is used. However, when IFN is added to TNF and DDP and EPI, there is promoting and vegetating effect on normal skeletal muscle cells in low concentration, in spite of inhibitory effect in high concentration.
