小麦体细胞无性系的建立及抗赤霉病突变体的诱导 被引量:7

摘要 利用小麦鄂恩 1号和 K6两个品种的胚轴、胚芽鞘作外植体建立了体细胞无性系 ,获得了可用于突变体诱导与筛选的胚性愈伤组织细胞 .结果表明 ,不同基因型和不同外植体在愈伤组织诱导率、愈伤组织生长状态以及愈伤组织分化上均有差异 ,且均受激素的调控 .赤霉菌粗毒素处理后 ,胚性愈伤组织在培养过程中随毒素浓度升高其生长受抑加剧 ,褐变增加 ,有的出现死亡 .诱变后的愈伤组织的分化力比对照降低 .诱变的再生植株经赤霉菌液感染后在培养中其生长状况优于对照 . This paper has presented the establishment of somatic cell clone by culturing the plumalar axis and embryonic bud sheath of wheat “E En1” and “K6”.The embryonic cell clones can be used in the induction and selection of mutant. The results show as follows: The growth,the induced percentage and the differentiation of calli are apparenthy different in different genotypes and different explantes, and they are adjusted to hormones.In culture,when the toxin concentraton increases,the embryonic calli growth becomes slow,the colour turns brown,and parts of them are dead.The differentiated percentage of calli treated with toxins becomes lower than that of CK calli which are not treated with toxins.The plantlets from induced calli when infected with scab can grow in media better than CK plantlets.
