摘要
目的观察Wnt1诱导分泌蛋白-1的表达,探讨其在结直肠腺瘤形成及癌变过程中的作用。方法用免疫组织化学LSAB法分别检测100例结直肠癌、58例结直肠腺瘤、40例正常结直肠黏膜中WISP-1蛋白的表达。结果结直肠癌、结直肠腺瘤、正常结直肠黏膜WISP-1表达阳性率分别是69.0%(69/100)、39.7%(23/58)和20.0%(8/40),组间差异有统计学意义(P<0.01),即在癌前病变开始逐渐出现高表达趋势。WISP-1的表达在结直肠癌中与分化程度、Dukes分期和淋巴结转移均密切相关(P<0.05);WISP-1的表达在结直肠腺瘤中与腺瘤大小,不典型增生程度、数目及组织学类型有关(P<0.05)。结论WISP-1可能是结直肠肿瘤发生发展中的重要促进因子,参与了腺瘤早期形成以及癌变过程,其可以作为结直肠癌早期诊断的标志和治疗的新靶点。
To explore the possible role of Wnt-1 induced secreted protein-1 (WISP-1 ) in colorectal adenoma neoplasty and cancerization by analyzing the expression of WISP-1. Methods By using immunohistochemical LSAB technique, the WISP-1 expression was detected in 100 cases of colorectal cancers, 58 cases of colorectal adenomas and 40 cases of normal colorectal mucosa. Results The positive rate of WISP-1 expression in colorectal carcinoma, adenoma and normal mucosa was 69.0% (69/ 100), 39.7% (23/58) and 20.0% (8/40) respectively with the difference being significant among them (P 〈 0.01 ). The expression of WISP-1 was increased significantly in the premalignant lesion tissue. Significant relationship was observed between WISP-1 expression and adenoma number, size, histological shape, degree of atypical hyperplasia in colorectal adenoma (P 〈 0.05 ). WISP-1 expression was closely related to histological grade, Dukes staging, and metastasis to lymph node in colorectal cancer (P 〈 0.05). Conclusion WISP-1, as a stimulative factor, participates in adenoma neoplasty and tumorigenesis. WISP-1 may be selected as a specific early diagnosis marker and therapy target in colorectal cancer.
