摘要
目的评价自行研制小儿胸腔镜心脏手术各种动、静脉插管的细胞毒性。方法对插管材料进行体外细胞培养,通过琼脂覆盖实验及噻唑蓝(MTT)实验观察细胞存活情况及活性,并与进口同类产品相对照。结果琼脂覆盖实验结果显示细胞溶解率小于20%,与对照组之间差异无统计学意义;噻唑蓝(MTT)结果显示50%材料浸提液1、3、5、7 d的细胞相对增殖率分别为85%、83%、81%及78%,具有轻微的细胞毒性。结论自行研制插管材料具有良好的生物相容性,细胞毒性轻微,可进一步应用于临床。
To evaluate the cytotoxicity of the PVC materials used in the modified cannulae in thoracoscopic assisted pediatric cardiac surgery. Methods Through in vitro cell culture, agar overlay test and MTT assay, cytotoxicity and cell activity of the self-developed PVC materials were evaluated and compared with controls. Results Agar overlay test showed that the cytolysis rate of the self-devel- oped PVC materials was lower than 20% , and there was no statistical difference with control group. MTT assay results revealed that the relative growth rate (RGR) of 50% leaching liquid after 1,3,5 and 7 days was 85% , 83% , 81% and 78% respectively. The self-developed PVC materials had slight cytotoxicity as compared with Medtronic DLP cannulae. Conclusion The PVC materials used in the modified arterial cannulae in thoracoscopic assisted pediatric cardiac surgery have little cytotoxicity and good biocompatibility. The newly designed cannulae set can be further applied to clinical use.
