摘要
采用高离子强度法分离出不同细胞质雄性不育胞质油菜叶绿体,裂解制备并纯化叶绿体DNA,限制性酶切分析比较了不同材料的ctDNA限制酶谱。结果表明,利用EcoRI酶切4种胞质叶绿体DNA,产生的DNA限制片断分子量在7.0~3.0kb之间,有明显的差异。
Rapeseed chloroplasts of 4 CMS materials were isolated by using high ionic strength with Bookjan's method. RNase was used to purify ctDNA from chloroplasts lysed by adding sarkosyl and proteinase K. EcoRI endonucleic restriction patterns of ctDNA reveals significant differences existed among the 4 CMS (polima CMS. nap CMS、ogu CMS and Ogu CMS mutant).
