橄榄降脂胶囊对高脂血症大鼠血脂及其肝脏低密度脂蛋白受体基因表达的影响

Effect of Ganlan Jiangzhi capsule on gene expression of liver LDLR and blood fat in rat with hyperlipoidemia

目的:探讨中药橄榄降脂胶囊的降脂机制,为临床应用提供实验依据。方法:采用酶法检测大鼠血清总胆固醇(TC)、甘油三酯(TG);采用磷钨酸-镁沉淀法检测HDL-c、LDL-c,RT-PCR法测定肝低密度脂蛋白受体(LDLR)基因表达。结果:模型组大鼠TC、TG、LDL-c均显著高于正常组,HDL-c显著低于正常组(P<0.05)。与正常组比较,模型组大鼠肝LDLR mRNA表达降低,P<0.05。各治疗组肝LDLR mRNA表达升高,与模型组比较P<0.05。结论:橄榄降脂胶囊可能通过调节肝LDLR mRNA表达而纠正脂质代谢紊乱。

Objective： To discuss the mechanism of Ganlan Jiangzhi Capsule in regulating blood fat and provide scientific evidence for its clinical application.Methods： The expression of TC and TG in blood serum of rats were determined by enzymic method;expression l of HDL-c and LDL-c were determined by PTA-Mg^2＋ precipitation method;LDLR gene expression level in liver of rat were analyzed by RT-PCR.Results： Compared with the normal group,TC,TG,LDC-C expression in model group were increased significantly,liver LDLR mRNA and HDL-c expression reduced.Compared with the module group,liver LDLR mRNA expression was increased in the treatment group.Conclusion： Ganlan Jiangzhi Capsule could improve lipid metabolic disorder by regulating expression of liver LDLR mRNA.
