摘要
目的探讨己酮可可碱对角质形成细胞的影响及其用于糖尿病足溃疡治疗的可能机制。方法对糖尿病足患者下肢手术后剩余皮肤,采用皮肤器官培养法(浮游法)培养皮肤标本,观察己酮可可碱对角质形成细胞游走的影响。结果浓度为3×10^(-5)mg/mL的己酮可可碱,对皮肤角质形成细胞游走有显著的促进作用。经过120小时的培养,己酮可可碱其他各个浓度组角质形成细胞游走距离均略高于对照组,但未见具有统计学意义的显著性差异。结论一定浓度的己酮可可碱具有促角质形成细胞游走的作用,且可能是其治疗糖尿病足溃疡的作用机制。
Objective To investigate the mechanism of pentoxinfyline on ulcerous healing of diabetic foot. Methods Use skin culture technique to cultivate the skin explants of patients with diabetic foot and observe the effect of varying concentrations of pentoxinfyline on migration of keratinocytes. Results Keratinocyte migration could be promoted by pentoxinfyline, especially at the concentration of 3×10^-5mg/mL, whereas there was no change seen with pentoxinfyline at remainder concentration. Conclusion Pronlption of the migration of keratinocytes of diabetic foot may be the mechanism of pentoxinfyline at appropriate concentration on ulcerous healing of diabetic foot.
