摘要
目的研究槲皮素对AlCl3致衰老模型小鼠记忆障碍的保护作用及其机制分析。方法以AlCl3(60 mg/kg皮下注射7 d)造成小鼠记忆障碍模型,采用小鼠避暗试验和跳台试验,观察槲皮素(20、10、5 mg/kg灌胃15 d)对记忆障碍模型小鼠的保护作用,并于第16天对各剂量组小鼠进行脑组织超氧化物歧化酶(SOD)活性及血浆和脑组织谷胱甘肽过氧化物酶(GSH-PX)活性测定。结果槲皮素各个剂量组均能明显改善AlCl3所致被动回避性记忆障碍,增加SOD和GSH-PX活性。结论槲皮素对AlCl3致衰老小鼠记忆障碍有保护作用,其作用机制可能是通过增强抗氧化酶GSH-PX和SOD活性,清除氧自由基对中枢神经系统神经细胞的损伤。
Objective To study the protective' effect and mechanism of quercetin on dysmnesia of acute senile mouse model induced by AlCl3. Methods After subcutaneous injection of AlCl3 60 mg·kg^-1 for 7days and intragastric administration of quercetin 20, 10 and 5mg·kg^-1 for 15days, the effect of quercetin on learning and memory were observed by dark avoiding test and step-down test, and the superoxide dismutase (SOD) activities in cerebrum and glutathione peroxidase (GSH-PX)activities in plasma and cerebrum were measured. Results Quercetin 20,0 and 5mg·kg^-1 significantly improved memory impairment induced by AICI3 and enhanced the activities of GSH-PX and SOD. Conclusion Quercetin showed protective effect on brain memory impairment in mice with AlCl3 induced acute insenecence. The mechanism perhaps involve enhancing the activities of GSH-PX and SOD, eliminating free radicals, protecting the brain neuron from the harm of lipoperoxide.
