摘要
目的观察去卵巢后痴呆大鼠模型海马CA1区β淀粉样蛋白(Aβ)及tau蛋白阳性神经元的变化及倍美力(Premain)对其的影响,探讨倍美力的脑保护作用机制。方法采用穹窿海马伞切除术制备AD模型鼠,切除穹窿海马伞、双侧卵巢切除制备痴呆及雌激素缺乏复合模型鼠,采用Morris水迷宫观察其行为学改变,利用免疫组织化学染色法测定大鼠海马CA1区Aβ及tau蛋白阳性神经元的表达。结果倍美力治疗组大鼠学习、记忆能力增强,海马CA1区Aβ及tau蛋白阳性神经元的表达低于正常组及模型组。结论倍美力有助于改善痴呆及雌激素缺乏复合模型鼠的学习及记忆能力。
Objective To observe the changes of positive cells of amyloid beta (Aβ) and tau protein in hippocampus CA1 region of the ovariectomized dementia model rats and the effect of Premain on it to discuss the action mechanism of protection of Premain on brain. Methods The dementia and hormonoprivia model rats were established by ovariectomy and transected hippocampal fimbria-fornix. The ability of learning and memory were tested with Morris water maze and the positive cells of Aβ and tau protein in hippocampus CA1 re- gion were measured with immunohistochemistry. Results The ability of learning and memory were increased obviously in Premain group. The expression of Aβ and tau protein was decreased significantly in Premain group compared with those of model and control groups. Conclusions Premain could improve the ability of learning and memory by decreasing the level of Aβ and tau protein in hippocampus CA1 region in dementia and hormonoprivia model rats.
