去痴灵对AD模型小鼠脑内β-淀粉样前体蛋白和脑啡肽酶基因表达的影响 被引量:4

摘要 目的探讨去痴灵对老年性痴呆(Alzheimer′s disease,AD)小鼠学习记忆及脑内β-淀粉样前体蛋白(amyloid beta-protein precursor,APP)和脑啡肽酶(Neprilysin,NEP)基因表达的影响。方法选用昆明种小鼠,腹腔注射D-半乳糖和灌胃三氯化铝制备AD模型小鼠,第51天开始,治疗组分别灌胃低、中、高剂量(10、20、40g.kg-1.d-1)的去痴灵,以石杉碱甲作为阳性对照药,连续40d。通过Morris水迷宫和半定量反转录聚合酶链反应(RT-PCR)测定去痴灵对AD小鼠学习记忆和脑内APP和NEPmRNA含量的影响。结果低、中、高剂量的去痴灵均能改善AD小鼠的学习记忆能力,下调APP基因表达,高剂量去痴灵能上调NEP基因的表达。结论去痴灵能改善AD小鼠学习记忆能力,降低APP基因表达及增加NEP基因表达。 Objective To observe the effect of Quchiling on the capacity of learning and memory, beta-amyloid precursor protein (APP) and enkephalinase neprilysin (NEP) expression in the Alzheimer's disease (AD) model mice. Methods The Kunming mice were treated with D-galactose (D-gal) and A1C13 to make the AD animal model. The therapeutic groups were treated with high, moderate and low dosage of Quchiling (i. g. , 40 d) respectively since the 51st day. Huperzine A was made to be a positive control drug, The capacity of learning and memory was tested on all mice with Morris water maze, the mRNA content of APP and NEP in brain tissue was assayed by reverse transcription polymerase chain reaction. Results Quchiling can improve the capacity of learning and memory and reduce the mRNA content of APP. The mRNA content of NEP could be increased by high dose of Quchiling. Conclusions Quchiling could improve the capacity of learning and memory, down regulate the expression of APP and up-regulate the expression of NEP gene.
