摘要
应用酚/氯仿抽提法提取缢蛏基因组DNA,并以所提取的基因组DNA为材料进行酶切反应及RAPD分析。结果显示,用酚/仿提取法提取的缢蛏基因组DNA质量高、稳定性好,酶切效果明显,RAPD扩增条带清晰,能满足DNA的酶切、PCR和RAPD等分子生物学实验对于模板DNA质量高的要求。
The genomic DNA of Sinonovacula constricta was successfully extracted by the hydroxybenzene/chlorof orm method,and it was used in restriction digestion of DNA and RAPD analysis.The genomic DNA extracted by the hydroxybenzene/chloroform method was of high quality、well stability、distinct effect of enzyme cutting,and the bands of RAPD were clear.In addition,the method can satisfy the demand of high quality template DNA used in molecular biology experiments such as the restriction digestion of DNA,PCR,RAPD and so on.
