摘要
目的:观察胰岛素样生长因子-1(IGF-1)基因转染的骨髓基质细胞(MSCs)合成IGF-1、骨粘连素(osteone-ctin,ON)和Ⅰ型胶原(Collagen-I)的变化,进一步了解基因转染骨髓基质细胞的生物学特性。方法:将含有IGF-1基因的真核质粒转染入大鼠骨髓基质细胞(MSCs)中,以正常培养的MSCs为对照组,分别于1、3、5、7d对MSCs进行IGF-1、ON和Collagen-I免疫组化观察。通过图像分析检测IGF-1基因转染对MSCs合成IGF-1、ON和Collagen-I的影响。结果:IGF-1基因转染后可促进MSCs合成IGF-1、ON和Collagen-(Ip<0.05),第3d合成IGF-1、ON和Collage-I作用最明显。结论:IGF-1基因转染的MSCs较未转染细胞合成IGF-1、ON和Collagen-I显著增加。
Objective: To observe the expression level of IGF-1, ON, and collagen-1 secreted by IGF-1 gene transfected MSCs,and further understand the biological characteristics of MSCs. Method: Transfect rat MSCs with eukaryon plasmid containing IGF-1 gene, and observe the expession level of IGF-1,ON,and collagen-1 on the day of lth, 2th,3th,5th,7th , respectively, with the method of immunohistochemistry (IHC) ,and the untransfected MSCs are considered as the control. The results are interpreted by image analysis. Result. IGF-Transfected MSCs can promote the higher level of IGF-1, ON and collagen-1 (P〈0.05) ,and the best effect can be observed on the 3th day. Conclusion: The significantly increased level of IGF-1, ON, and collagen-1 can be promoted by IGF-1 transfected MSCs.
