奥硝唑缓释膜体外抑菌性及体内释放度的研究 被引量:4

摘要 目的:研究以羧甲基壳聚糖为载体的奥硝唑缓释膜(药膜,下同)的体外抑菌性及体内释放度。方法①.固体培养基体外抑菌法观察药膜对变形链球菌和具核梭杆菌的抑菌性。②.12只大鼠,钝性分离牙龈制备牙周袋模型,分别给予腹腔注射奥硝唑和牙周袋放置药膜,腹腔组于给药后0.5h,1h,2h,3h,4h,牙周袋组于24h,48h,5d取龈沟液测药物含量。结果:①.药膜对所测致病菌有良好的抑菌性。②.腹腔注射药物剂量是局部置药的140倍,而龈沟液药物浓度是局部置药的1%~5%。结论:药膜有良好的抑菌性,能在牙周袋内较长时间维持有效浓度,是一种既速效又能缓释的新剂型。 Objective: To evaluate in Vitro antibacterial activities and the in vivo release rate of the ornidazole slow- release membranes. Method: ① .The in vitro antibacterial activity against Streptococcus mutans and Fusobacterium nucleatum was observed on solid culture medium. ②. 12 Wistar rats whose gingivae were bluntly separated from the teeth to establish the periodontal pocket models. The animals were respectively treated with abdominal injection of ornidazole (A- group) and local application of the ornidazole slow release membranes (L-group). Gingival crevicular fluid (GCF) was gained at 0.5, 1,2,3,4 h in A-group and 24 h, 48 h, 5 d in L-group to examine the concentration of ornidazole. Result: ①. It was also found that the membrane had effective antibacterial action against Streptococcus mutans and Fusobacterium nucleatum. ②. The dosage of ornidazole in A-group was 140 times of that in L-group while the ornidazole concentration of GCF in A-group is 1%-5% of that in L-group. Conclusion: The membrane possesses favorable antibacterial activities. It can maintain therapeutic concentrations within the periodontal pocket for a long period of time. So it can be used as a new slow-release delivery system in the treatment of periodontitis.
