利用顶空SPME-GC-MS联用技术检测苹果汁中嗜酸耐热菌代谢产物的研究 被引量:9

摘要 以苹果汁中分离获得的1株嗜酸耐热菌和标准嗜酸耐热菌为材料,采用优化的AAM培养基进行培养,用顶空SPME-GC-MS联用技术对其代谢产物进行了研究。结果表明:①嗜酸耐热菌的主要代谢产物为愈创木酚和2,6-二溴苯酚;②单因素分析发现愈创木酚含量远远高于2,6-二溴苯酚,可以作为整个苹果汁中嗜酸耐热菌污染程度和风味败坏的衡量指标;③正交试验分析得到顶空固相微萃取法萃取愈创木酚的条件为DVB/CAR/PDMS萃取头,添加1.8gNa2SO4在温度55℃下萃取20min。 Use a local alicyclobacillus acidoterrestris which isolated from the fruit juice processing company for the analysis. The optimized AAM broth was used to incubate the alicyclobacillus acidoterrestris, using headspace SPME-GC-MS technical to detect the metabolite. The results showed that: 1. The main metabolites of Alicyclobacillus acidoterrestris were guaiacol and 2,6-dibromophenol; 2. Single factor anlysis shows that the yield of guaiacol was much more than the 2,6-dibromophenol, and indict.ted the spoilage caused by alicyclobacillus acidoterrestris in apple juice; 3. The optimized conditions based on orthogonal experiment were: DVB/CAR/PDMS fibre, 1.8g NaSO4 at 55℃ for 20min extraction.
