摘要
探讨了麸皮、秸杆粉、豆饼等碳氮源及含水量对产酶的影响.在单因素实验的基础上,采用Box-Behnken实验设计对黑曲霉产酸性α-淀粉酶的培养基进一步优化,实验结果运用SAS(8.0版)统计软件进行统计分析,得到最佳的产α-淀粉酶的培养条件为:76%麸皮、12%玉米淀粉、10.5%豆饼、1.5%NH4Cl、料水比为1∶1.在最佳的培养基上于30℃发酵72 h,产酶达198.4 U/g,α-淀粉酶的最佳作用pH为4-4.5.
The effects of nutrition ingredient and water addition on solid fermentation producing acidic amylase with Aspergillus niger was investigated. Box-Behnken Experimental design (BBD) was used to optimize fermentation medium. The best producing acidic amylase medium contained 76% wheat bran, 12.5Mcorn starch, 10.5%soybean flour,1.5 % NH4Cl and the ratio of water was 1 :1. In optimal condition, the yield of amylase is 198.4 U/g. and the amylase showed a high stability at pH4-4.5.
