RNA干扰基因敲低雌激素受体β对雌激素诱导人成骨样细胞护骨素表达的影响 被引量:2

Effect of estrogen receptor β knocked down by RNA interference on osteoprotegerin expression in estrogeninduced human osteoblast-like cells

摘要 RNA干扰敲低人成骨样细胞株MG63中雌激素受体β后,RT-PCR法发现不同浓度的17β-雌二醇均能上调MG63细胞中的护骨素mRNA表达水平,其中以10^(-7)mol/L的作用最强。而这种作用不能被G蛋白抑制剂suramin所抑制。提示雌激素受体β可能不涉及雌激素上调MG63细胞护骨素表达的调控作用。 [Summary] After-estrogen receptor β (ERβ) in MG63 cell was knocked down by RNA interference, expression of osteoprotegerin (OPG) induced by 17β-estradiol was assayed by RT-PCR. 17β-estradiol with various concentration obviously upregulated the expression of OPG mRNA of MG63 cell with the maximal effect at the concentration 10^-7 mol/L, which was not inhibited by suramin (G protein inhibitor). The present result suggested that ERβ was not involved in regulation of OPG mRNA expression in MG63 cell by 17β-estradiol.
