摘要
目的:观察胶原在不同的预冻温度下其内部孔径大小的情况。方法:将牛I型胶原粉末在0.5M的醋酸溶液中溶解后离心,把加入胶原的6孔板分别放入-20℃、-70℃冰箱过夜(约12h),预冻后胶原置于-70℃冻干机内冻干48h,扫描电镜(SEM)测量胶原支架内部孔径。结果:胶原溶液经冷冻干燥后呈膜状结构,经SEM测量-20℃预冻温度所得胶原侧面孔径大小在107.21~215.49μm之间,-70℃预冻温度所得胶原侧面孔径大小在62.18~133.58μm之间。结论:不同的预冻温度所形成的胶原海绵内部孔径大小不同,胶原支架在-70℃的预冻温度下获得了理想的孔径大小。
Objective: To investigate the pore size of collagen sponge at different prefreezing temperature. Methods: Collagen sponge was dissolved in 0.5 M acetic acid and centrifugalized. Collagen solution was poured into 6 hole planks and kept at - 20℃ and - 70℃ for 12 hours, which was stored in - 70℃ freezer for 48 hours. The pore size of collagen scaffold was measured by scanning electron microscope (SEM). Results: Collagen sponge ap- peared as white film, fibrous, connected and formed pore. The pore size of collagen scaffold freezed at - 20℃ ranged from 107.21 to 215.49 tun under SEM, while at - 70℃ the pore size of collagen scaffold ranged from 62.18 to 133.58 μm. Conclusion: The pore side of collagen sponge depends on pre-freezing temperature. - 70℃ pre- freezing temperature is suitable to collagen sponge to obtain ideal pore side.
