摘要
目的研究低温冻存对处周血单个核细胞(PBMNCs)诱导为CIK(cytokine-inducedkiller)细胞及抗肿瘤效应的影响。方法采用非程控降温方法冻存外周血单个核细胞,复苏后诱导为CIK细胞,与新鲜的未经冻存的外周血单个核细胞诱导的CIK细胞进行扩增能力、细胞纯度、细胞毒活性等比较。结果冻存后的外周血单个核细胞诱导生成的CIK细胞在扩增能力、细胞纯度、细胞毒活性等方面与新鲜细胞直接诱导生成的CIK细胞无显著性差异。结论冻存后的外周血单个核细胞可以诱导生成具有抗肿瘤活性的CIK细胞,研究结果对CIK细胞的应用具有重要意义。
Objective To explore the anti-tumor effect of CIK cells induced with cryopreserved peripheral blood mononuclear cells (PBMNCs). Methods To compare the proliferative ability, purity and cytotoxicity between the CIK cells induced with cryopresevecl FBMNCs and the CIK cells from the fresh PBMNCs. Results The CIK cells induced with cryopreserved PBMNCs showed no significant difference from those induced with fresh PBMNCs in proliferative ability, purity and cytotoxicity. Conclusion This study demonstrates that human CIK cells induced with cryopreserved PBMNCs maintain their anti-tumor Ability. These finding have important implications for the application of these cells as adoptive cellular therapy.
