用原位杂交技术检测人子宫颈癌组织中HPV16E_6转化基因的研究

摘要 应用地高辛非放射性标记与检测系统制备人乳头瘤病毒16型E6(HPV16E6)转化基因DNA探针,经用斑点杂交检测,探针特异性强,灵敏度达0.1pg,应用组织切片原位杂交技术检测了44例子宫颈癌组织。结果表明,HPV16E6阳性27例、占61.36%,5例正常对照子宫颈组织中未检出。提示HPV16E6转化基因与子宫颈癌密切相关,为阐明子宫颈癌的病因和HPV16E6在诊治子宫颈癌中的作用提供了理论依据。 The relationship between human papillomavirus type16 E 6 transforming gene and human cervical carcinoma was performed.Human PV16E 6 DNA fragment was labelled using Digoxingen labelling and detection kit,the specificity and sensitivity was detected by heterologous and homologous dot blot hybridization,its sensitivity was 0.1 pg.Forty four human cervical carcinoma samples were screened for the presence of HPV16E 6 transforming gene in formalin fixed paraffin embedded cervical carcinoma tissue by in situ hybridization with HPV16 E 6 probe.The positive result of HPV16 E 6 gene was detected in 27 of 44 cases(61.36%).The result indicated HPV16E 6 transforming gene may play an important role in carcinogenesis of human cervix.The in situ hybridization method was a value technique in detecting HPV infection.
