摘要
建立检测沙眼衣原体的PCR方法,计算机辅助设计的引物位于编码沙眼衣原体主要外膜蛋白(MOMP)的基因区内,扩增产物为144bp。该引物具有高度的特异性和敏感性,其最小检出量为5fg。应用该引物检测了52份宫外孕组织中沙眼衣原体DNA,测得宫外孕患者沙眼衣原体的阳性率为40.4%,明显高于对照组的12.5%(P<0.01)。
The PCR assay for detection of chlamydia trachomatis (CT) DNA was developed.The oligonucleotied primer designed with computer was located at major outer membrane protein gene sequence of chlamydia trachomatis,it's amplified products was 144bp.The CT PCR assay had high specificity and sensitivity,the 5fg CT DNA could be detected.Using this primer,CT DNA in 52 salpingian tissues of ectopic pregnancy was detected to study the relationship between ectopic pregnancy and chlamydia trachomatis,and to compare with traditional cell culture.The positive percentage of ectopic pregnancy patients was 40.4%(21/52) which was significantly higher than that of control group(12.5%).It is first proved that chlamydia trachomatis is present in ectopic pregnancy tissues by DNA detection.
