摘要 目的:探讨外源性p27Kip1基因对人乳腺癌MCF-7细胞株细胞周期和增殖的影响。方法:应用脂质体转染法将含人野生型p27Kip1基因质粒DNA转染乳腺癌MCF-7细胞,免疫细胞荧光化学方法验证p27Kip1基因转染MCF-7细胞后蛋白表达情况;流式细胞仪检测细胞周期的变化;细胞计数法绘制细胞生长曲线,观察外源性p27Kip1对乳腺癌细胞MCF-7生长的影响。结果:免疫细胞荧光化学方法证实p27Kip1基因转染MCF-7细胞后存在蛋白表达;流式细胞仪检测结果提示,细胞周期出现G0/G1期阻滞;细胞计数法绘制细胞生长曲线显示细胞增殖明显抑制。结论:外源性p27Kip1基因对乳腺癌MCF-7细胞生长和细胞周期有明显抑制作用。 Objective: To explore the effects of exogenous p27kipl gene on the growth and cell cycle of MCF-7 cells. Methods : The p27kipl cDNA was cloned into expressor vector pcDNA3.1 and transfected into human breast cancer cell line MCF-7 with lipofectamine. The expression of p27kipl protein in MCF-7 cells were examined by immunoflurescence, and its effects on the cell growth and cell cycle were observed by FACS. Results: Over-expression of p27Kipl protein was observed in the transfected cells, and the proliferation of MCF-7 cells was greatly inhibited and the cell cycle was arrested in the G0/Gl phase after exogenous p27KiPlexpression. Conclusion: Exogenous p27Kipl gene can inhibit the growth and cell cycle of human breast cancer cell line MCF-7.
