大黄素对人增生性瘢痕成纤维细胞的增殖及细胞周期作用的实验研究 被引量:1

摘要 目的观察大黄素对体外培养的人增生性瘢痕成纤维细胞增殖及细胞周期的作用,以寻找治疗人增生性瘢痕的有效药物。方法将切取于增生性瘢痕患者的瘢痕组织标本分成A(对照组)、B、C、D四组,每组6个标本,行体外成纤维细胞培养,采用0、50、100、200μg/ml的大黄素依次对A、B、C、D组的成纤维细胞进行干预,应用MTT比色法和流式细胞仪检测细胞增殖和细胞周期的变化。结果B、C、D组标本中成纤维细胞的增殖受到抑制,抑制于G0/G1期(P〈0.05),与A组比较,差异有统计学意义(P〈0.05)。而且大黄素在50~200μg/ml的范围内,对成纤维细胞增殖的抑制有浓度的依赖性。结论大黄素具有体外抗纤维化作用,可能成为治疗人增生性瘢痕的有效药物。 Objective To observe the effects of emodin on the proliferation and cell cycle of the cultured human hypertrophic scar fibroblasts in vitro and seek the effective drug for human hypertrophic scar. Methods The scar tissue specimens incised from patients with hypertrophic scar were divided into four groups: A(control group), B, C, D. Each group containd six specimens. The fibroblasts were cultured in vitro. Emodin in various concentrations(0, 50 μg/ml, 100 μg/ml, 200 μg/ml) were added to the medium. The MTT test and flow cytometry medxxts were used to detect the effects of emodin on the proliferation and cell cycle of the cultured human hypertrophic scar fibroblasts. Results The proliferation of fibroblasts in the B, C, D group was inhibited at G0/G1 period (P 〈 0.05 ), and the difference was significant when compared with A group (P 〈 0.05). The inhibition of Emodin on the fibroblasts had concentration dependence during the concentration of 50 - 200μml. Conclusion Ernodin has anti - fibrotic effects in vitro and may be an effective drug for human hypertrophic scar.
