扶正解毒颗粒拮抗硫酸镍诱发大鼠睾丸某些生化指标的变化

摘要 目的探讨中药扶正解毒颗粒(FJK)对硫酸镍(NiSO4)致大鼠睾丸某些生化指标变化的拮抗作用。方法40只Wister雄性大鼠随机分为5组:生理氯化钠(NS)组;NiSO42.5mg/kg染毒组,NiSO4连续染毒30d;FJK10.0、5.0、2.5g/kg3个处理组,3组NiSO4染毒方法和剂量同染毒组,于第10天同时灌胃给予FJK处理。各组均于第31天处死大鼠,制备睾丸组织匀浆并提取线粒体。结果睾丸组织匀浆中NS组和NiSO4组谷胱甘肽过氧化物酶分别为(40.88±7.24)和(18.20±4.24)×103U/g,总抗氧化能力为(6.38±0.84)和(2.60±0.82)×103U/g,脂质过氧化物为(1.61±0.33)和(3.07±0.28)mol/g,活性氧为(5.36±2.37)和(9.71±1.62)×103U/g,2组间各项指标差异有非常显著性(P<0.01);经FJK拮抗处理后,3个FJK组上述各项指标的NiSO4毒作用均显示减轻(P<0.05或0.01)。各组大鼠睾丸细胞线粒体中总抗氧化能力、脂质过氧化物和活性氧水平的变化情况与睾丸组织匀浆相似。结论中药FJK对Ni-SO4所诱发的谷胱甘肽过氧化物酶活力、总抗氧化能力、脂质过氧化物和活性氧含量异常变化具有逆转作用。 Objective To explore the antagonistic effect on certain biochemical changes in rat testis induced by nickel sulfate (NiSO4) with a Chinese herbal drug Fuzhengjiedu granule (FJK). Methods 40 Wister male rats were divided into five groups: a cnntrol group (NS), a NiSO4 intraperitoneal injection group at the dose of 2.5 mg/kg NiSO4 for 30 days, 3 FJK treated groups at the doses of 2.5, 5.0 and 10. 0 g/kg FJK respectively. All treated groups were given FJK by gavage daily from the tenth day of NiSO4 treatment. Animals were executed at the thirty-first day, homogenate were prepared and chondriosome were extracted. Activities of glutathione peroxidase ( GSH-Px), total antioxidation capability ( T-AOC ), contents of lipid peroxide (LPO) and reactive oxygen species (ROS) in testis tissues were detected. Results Respectively in NS and NiSO4 groups, GSH-Px were(40. 88 ±7. 24) and( 18. 20 ±4. 24) × 103 U/g, T-AOC were(6. 38 ±0. 84) and(2. 60 ±0. 82) × 10^3 U/g, LPO were( 1.61 ± 0. 33) and (3.07 ± 0. 28)mol/g, ROS were (5. 36 ± 2. 37 )and (9. 71 ± 1.62) × 10^3 U/g in testis homogenate, showing very significant differences( P 〈 0. 01 ). After FJK treatment, the results showed that the toxic effect of NiSO4 in all three treated groups reduced, The contents of LPO, T-AOC and ROS in testis cell chondriosome were similar to those in testis homoge- hate. Condusion FJK could have a reverse effect on abnormal changes of GSH-Px activities and the contents of T-AOC, LPO and ROS induced by nickel sulfate.
