雷公藤甲素对变应性接触性皮炎小鼠淋巴细胞周期及凋亡的影响 被引量:3

Influences on the Cell Cycle and Apoptosis of T Lymphocyte in Mice Models of Allergic Contact Dermatitis Cultured in Vitro Induced by Triptolide

摘要 目的研究雷公藤甲素对变应性接触性皮炎小鼠淋巴细胞周期及凋亡的影响。方法以二硝基氯苯制作ACD小鼠模型,体外培养淋巴细胞,分别以0.1、1.0、10μmol/L浓度雷公藤甲素作用后,采用Annexin V-FITC试剂盒,PI染料流式细胞仪分别检测细胞周期及细胞凋亡。结果甲素三个浓度组对G1、S期细胞的影响与对照组比较差异均具有统计学意义(P<0.01),对G2期及亚二倍体细胞的影响只有高、中浓度组与对照组之间的差异具有统计学意义(P<0.01)。同时高浓度组对G1、S、G2和亚二倍体的影响与低浓度组比较差别也具有统计学意义(P<0.01)。三个浓度组凋亡细胞比率与对照组比较差异均有显著性(P<0.01);其中高浓度组凋亡细胞比率高于低浓度组及中浓度组(P<0.01),而中浓度组与低浓度组比较差异无统计学意义(P>0.05)。结论诱导体外培养的ACD小鼠淋巴细胞凋亡,调节淋巴细胞周期,可能是雷公藤甲素治疗ACD的机理之一。 Objective To research on the cell cycle and apoptosis of T lymphocyte in mice models of Allergic Contact Dermatitis (ACD) cultured in vitro induced by Triptolide. Methods The mice models of ACD were induced by dinitrochlorobenzene (DNCB),and cultured the cells in vitro for 48 hours. The drug level in the control groups was 0.0 μmol/L,and in Triptolide groups were 0.1,1.0 and 10 μmol/L. The cell cycle and apoptosis of T lymphocyte was detected by flow cytometry (FCM). Results The ratio of G1 and S-phase in all the Triptolide treatment group at different concentration compared with the control group had statistical significance,and the ratio of G2-phase in the 10μmol/L group was much lower than that in the 0.1 μmol/L group(P〈0.01). The ratio of apoptosis cells in all the triptolide treatment group at different concentration was much higher than that in the control group(P〈0.01),and the radio of the high concentration gruop was much higher than that in the 0.1 and 1 μmol/L groups(P〈0.01). Conclusion Triptolide could induce apoptosis in the ACD mice models T lymphocyte in vitro,and it was one of the possible mechanisms to depress the activated T lymphocyte.
