水稻花粉组织特异性启动子捕获实验 被引量:4

Experiment of pollen-specific promoter's trapping in rice

摘要 组织特异性启动子是细胞分化、基因特异性表达、基因工程研究及实际应用中的重要工具。一个由带有UidA基因而无启动子的pPLGUS改造成的质粒通过基因枪转化进水稻材料中,对转基因材料的多种不同组织进行了X-gluc显色检测GUS活性。对获得的15个独立转化株后代进行筛选,初步观察到其中多株GUS阳性。通过连续三代遗传分析,证明至少3株水稻花粉组织特异性启动子被成功捕获,为进一步研究应用奠定了基础。 The tissue specific promoter is an important element of cell differentiation, gene specific expression, genetic engineering and its application. A promoterless UidA containing plasmid derived from pPLGUS was transformed into rice ( Oryza sativa L. ). Histoehemical analysis of various tissues in transgenic rice was carried to examine tissue specific expression of GUS activity. By analyzing the acquired 15 transformants, a transformant line with pollen- specific GUS expression was successfully trapped. By 3 generations genetic study, there are 3 strains were determined to be pollen - specific promoter trapped. It is a good foundation for further study and application.
