rhG-CSF和rhGM-CSF动员小鼠外周血干细胞效果的比较研究 被引量:1

摘要 目的比较重组人粒细胞集落刺激因子(rhG-CSF)和重组人粒-巨噬细胞集落刺激因子(rhGM-CSF)单独、同时或序贯联合应用对小鼠外周血干细胞(PBSC)的动员作用,从而选择最佳的动员方案。方法72只BALB/c小鼠随机分成6组,每组12只。A组皮下注射rhG-CSF 400μg.kg-1.d-1;B组皮下注射rhGM-CSF 400μg.kg-1.d-1;C组皮下注射rhG-CSF 200μg.kg-1.d-1+rhGM-CSF 200μg.kg-1.d-1;D组皮下注射rhGM-CSF 400μg.kg-1.d-1(前3天)+rhG-CSF 400μg.kg-1.d-1(后2天);E组皮下注射rhG-CSF 400μg.kg-1.d-1(前3天)+rhGM-CSF 400μg.kg-1.d-1(后2天),连续注射5天。对照组连续5天皮下注射生理盐水0.2 ml。动态观察各组外周血白细胞计数、CD34+细胞比例、CFU-GM产率的变化。结果rhG-CSF与rhGM-CSF单独或联合使用时,外周血白细胞计数、CD34+细胞、CD34+CD38-细胞和粒-单核细胞集落形成单位(CFU-GM)产率明显高于对照组(P<0.01)。实验组中,含有rhG-CSF组的外周血白细胞计数、CD34+细胞产率明显高于单用rhGM-CSF组(P<0.01);含有rhGM-CSF组的CD34+CD38-细胞产率明显高于单用rhG-CSF组(P<0.01);rhG-CSF与rhGM-CSF联合组CFU-GM产率高于单用rhG-CSF、rhGM-CSF组(P<0.05)。结论rhG-CSF联合rhGM-CSF方案可作为外周血干细胞动员的一个较好的选择。 Objective To Optimize the regime for mobilizing peripheral blood stem ceils in mice by comparing the effects of recombinant human granulocyte colony stimulating factor ( rhG - CSF) and recombinant human granulocyte - macrophage colony stimulating factors ( rhGM - CSF) when used alone or in concurrent or sequential combination. Methods 72 BALB/c mice were randomly divided into 6 groups, 12 mice in each group. The experiment groups were assigned sequentially to the following 5 - day regimens for mobilization: rhG - CSF 400 μg · kg^-1· d^-1 alone; rhGM - CSF 400 μg· kg^-1 ·d^-1 alone; concurrent combination of rhG - CSF 200 μg · kg^-1 · d^-1 and rhGM - CSF 200 μg· kg^-1 · d^- 1 ; sequential combination of rhGM - CSF 400 μg· kg^-1 · d ^-1 for 3 days and rhG - CSF 400 μg · kg^-1 · d^ -1 for 2 days; sequential combination of rhG - CSF 400 μg· kg^- 1 · d ^- 1 for 3 days and rhGM - CSF 400 μg·kg^- 1 · d ^- 1 for 2 days. The control group was given a subcutaneous injection of saline 0.2 ml qd for 5 days. The white blood cell (WBC) count, the percentage of CD34 + ceils and the number of CFU - GM in peripheral blood of mice were dynamically observed. Results The WBC, CD34 + ceils, CD34 + CD38 - ceils and CFU - GM yield in peripheral blood of the experimental groups were all higher than that of the control group (P 〈0.01 ). The experimental groups treated with rhG - CSF alone or in combination with rhGM - CSF resulted in increase in number of WBC and CD34^+ cells in peripheral blood as compared to that of using rhGM - CSF alone ( P 〈 0.01 ). The number of CD34^+ CD38 - cell of the group treated with rhG - CSF alone was lower than that of the other experimental groups (P 〈0.01 ). The yield of CFU - GM was higher in the combination groups than in the single G- CSF and single rhGM -CSF groups (P 〈 0.01 ). Conclusions The combined use of G - CSF and GM - CSF is a better alternative for peripheral blood stem cell mobilization.
