高效液相色谱法测定替米沙坦的血药浓度

摘要 目的探讨HPLC法测定人血浆中替米沙坦浓度。方法XTerra RP18(4.6 mm×250 mm,5μm)为色谱柱,以甲醇-0.03 mol.L^-1磷酸二氢钾溶液(含0.6%三乙胺,用磷酸调pH至4.00)为流动相,流速0.8 ml.min^-1,检测波长298 nm,灵敏度2.000 AUFS,柱温30℃。结果替米沙坦的高、中、低3种浓度的相对回收率分别为109.7%、98.1%和101.2%,精密度日内RSD≤1.87%(n=5)、日间RSD≤5.14%(n=5);线性范围为24.8~1415.1μg.L-1,回归方程为:A=147.58c-1323.5(r=0.999 9,n=5);本方法的最低检测浓度为15μg.L^-1。结论该方法可用于替米沙坦临床血药浓度的监测。
