摘要
目的研究川芎嗪(tetramethylpyrazine,TMP)对癫痫的作用及其与神经递质谷氨酸(glutamate,Glu)、γ-氨基丁酸(γ-aminobutyric,GABA)的关系。方法采用腹腔注射戊四氮(pentylenetetrazol,PTZ)诱发大鼠癫痫模型。将SD大鼠随机分为3组,即:生理盐水对照组;戊四氮致痫组;川芎嗪+戊四氮致痫组。采用免疫组织化学方法检测各组大鼠大脑皮层、海马谷氨酸、γ-氨基丁酸阳性神经元的表达变化。结果戊四氮致痫组与对照组比,大脑皮层、海马Glu阳性神经元明显增多,大脑皮层、海马GABA阳性神经元明显减少,差异均有显著性意义(P<0.05)。川芎嗪干预后,大脑皮层、海马Glu阳性神经元明显减少,大脑皮层、海马GABA阳性神经元明显增多,差异均有显著性意义(P<0.05)。结论川芎嗪能上调GABA阳性神经元数目,下调Glu阳性神经元数目。川芎嗪可能通过抑制脑内Glu的产生,促进脑内GABA的产生,从而使神经元的兴奋性降低,抑制了癫痫的发生。
Objective To study the effects of tetramethylpyrazine(TMP) in treating epilepsy and its relation to the neurotransmitters of glutamate(Glu) and γ- aminobutyric(GABA) . Methods SD rats were selected and randomized into three groups: normal saline control group, pentylenetetrazol group and treatment of tetramethylpyrazine group. The changes of expression of Glu and GABA proteins in cortex and hippocampus of rats were detected by immunosuppressant method. Results The expression of the Glu proteins in pentylenetetrazol groups were significantly higher than those in saline control groups. While the expression of GABA proteins in pentylenetetrazol groups were significantly lower than those in saline control groups ( P 〈 0.05 ). The expression of the Glu proteins in those intreatment of tetramethylpyrazine groups were significantly lower than those in pentylenetetrazol groups. While the expression of GABA proteins in intreatment of tetramethylpyrazine groups were significantly higher than those in pentyle- netetrazol groups (P 〈 0. 05 ). Conclusion Tetramethylpyrazine can upregnlate the expression of GABA protein and downregnlated the expression of Glu. Tetramethylpyrazine maybe reduce the nerve excitation and inhibit the epilepsy through inhibiting the production of Glu and increasing the production of GABA in brain of rats.
