摘要
目的:优化知母甾体总皂苷的提取工艺。方法:以甾体菝葜皂苷元的提取率为指标,应用L9(34)正交试验设计优化了知母中甾体皂苷的纤维素酶法提取工艺。结果:影响酶法提取甾体皂苷的主次因素为:酶用量>提取温度>固液比>水解时间。纤维素酶处理能显著提高甾体皂苷的提取率,加酶后甾体菝葜皂苷元提取率的平均值为84.6%,而未加酶的结果则为58.1%。结论:酶法提取新工艺能显著提高知母甾体皂苷的提取率。
Objective: To optimize extractive technology for steroidal total sapogenin in anemarrhenae. Methods: To use extractive rate of steroidal sarsasapogenin as the target, the extract technology of cellulase method for steroidal total sapogenin in anemarrhenae was optimized with L9 (3^4) orthogonal experiment. Result: The sequence of factors to affect steroidal saponin extract as follow, dosage of enzyme 〉 extractive temperature 〉 ratio of solid and liquid 〉 hydrolytic time. The extract used cellulose could increase the extractive rate of steroidal saponin, the average value was 84.61% in extractive rate of steroidal steroidal sarsasapogenin after adding enzyme, that was 58.1% in that of no enzyme. Conclusion: The new technology extract with enzyme method can increase the extractive rate steroidal saponin in anemarrhenae obviously.
