摘要 目的:研究单磷酸腺苷(AMP)激活的蛋白激酶α2亚单位(AMPKα2)基因PRKAA2多态性与中国重庆地区汉族人群2型糖尿病(T2DM)的相关性。方法:选取中国重庆地区T2DM患者492例(病例组)、正常对照296例(对照组),采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)的方法,对PRKAA2第6内含子区单核苷酸多态性(SNP)位点rs857155(440A>C)进行基因分型。结果:PRKAA2多态性位点rs857155三种基因型AA、AC、CC频率在病例组中分别为0.161、0.567、0.272,在对照组中分别为0.192、0.586、0.223,差异无显著性(χ2=2.886,P>0.05);病例组和对照组等位基因频率分布差异亦无显著性(χ2=1.186,P>0.05)。结论:中国重庆地区汉族人群PRKAA2多态性位点rs857155与2型糖尿病无关联。 Objective:To study the relationship between 5'-AMP-activated protein kinase alpha-2 subunit gene PRKAA2 polymorphism and type 2 diabetes mellitus (T2DM) in Han population in Chongqing. Methods:In the study,492 patients with type 2 diabetes (Case group) and 296 normal control subjects (Control group) from Chongqing were enrolled. A single nucleotide polymorphism (SNP) rs857155 (440A〉C) in the intron 6 region of PRKAA2 was genotyped in all the subjects enrolled using PCR and restriction fragment length polymorphism (RFLP). Results:The frequencies of AA,AC and CC genotype in case group were 0.161,0.567 and 0.272 respectively,and 0.192,0.586 and 0.223 in control group.The distribution rates of genotype were found to be not significantly different between control group and case group (Χ^2=2.886,P〉0.05),and the allele frequencies also had no significient difference between two groups (Χ^2=1.186,P〉0.05). Concision:The polymorphism of rs857155 in PRKAA2 is not associated with T2DM in Han population from Chongqing .
