摘要
目的:探讨银杏叶提取物(EGb)对糖尿病大鼠睾丸和附睾组织活性氧及内皮素的影响。方法:Wistar雄性大鼠40只,随机分成三组,对照组(C),糖尿病组(DM组)和银杏叶(EGb)治疗组.应用链脲佐菌素(Streptozocin STZ)加高脂高糖饮食制备2型糖尿病模型,治疗组同时给予银杏叶提取物(EGb)10周后,取双侧睾丸和附睾组织检测组织中MDA含量、内皮素(ET)含量和SOD、GSH-Px活性。结果:2型糖尿病模型制备成功,糖尿病组与对照组比较,SOD、GSH-Px活性降低,差异有显著性意义(P<0.01),MDA含量和ET含量明显高于对照组(P<0.01)。而治疗组与糖尿病组比较,SOD、GSH-Px活性升高差异有显著性意义(P<0.01),MDA含量和ET含量明显低于糖尿病组(P<0.01)。结论:糖尿病活性氧(ROS)和ET含量增多,抗氧化酶(SOD、GSH-Px)活性下降,可能是造成睾丸、附睾细胞损伤,导致睾丸功能紊乱,进而导致性与生殖功能障碍的原因,而银杏叶提取物能有效降低MDA、ET的含量对糖尿病大鼠生殖系统的毒性损伤,增强SOD、GSH-Px活性,可见银杏叶提取物对糖尿病大鼠睾丸和附睾组织具有保护作用。
Objective: To observe the effects of extract Gingko biloba on ROS of testis and epididymis and ET of diabetic rats. Methods:Forty male Wistar rats were divided randomly into three groups, control group , diabetic group and EGb - treated group . we established type Ⅱ diabetes model with streptozotocin (STZ) and high lipid , high glucose diet and EGb -treated groups were given EGb simultaneously , after ten weeks ,the testis and epididymis were taken out for measurement of the levels of MDA content and ET content and activities of SOD and GSH - Px . Results:The type II diabetes model was prepared successfully and compared with the control group , SOD and GSH - Px activities dropped sharply with significant difference ( P 〈 0.01 ), MDA and ET content obviously increased (P 〈0.01 ),compared with the diabetic group ,SOD and GSH -Px activities increased obviously (P 〈0. 01 ), contents of MDA and ET evidently decreased ( P 〈 0.01 ) in EGb - treated group . Conclusion : Increased ROS and ET decreased antioxidant enzymes (SOD and GSH -Px) activities might give raise to testis and epididymis damage and their dysfunction ,which result in sexal and reproductive malfunction ,EGb can reduce MDA and ET content which cause toxic damage to diabetic rats reproductive system and increase the activities of 1SOD and GSH -Px ,EGb can protect the testis and epididymis of diabetic rats .
