摘要 目的:研究Nucleostemin特异性RNA干扰(RNAi)联合4种化疗药物对人食管癌细胞株Eca-109体外增殖的影响。方法:1.用NS特异性RNAi表达载体转染Eca-109细胞。2.实验分组:分为silencer组(S组)、normal组(N组)、单纯用药组和联合用药组。S组为转染NS特异性siRNA表达载体的Eca-109细胞;N组为正常Eca-109细胞;单纯用药组为单纯应用4种化疗药物作用于Eca-109细胞;联合用药组为转染NS特异性RNAi表达载体分别联合4种化疗药物作用于Eca-109细胞。3.利用细胞计数法、镜下细胞形态学观察和MTT法检测各组细胞的增殖情况。结果:联合用药各组增殖速率明显低于单纯用药各组,而增殖抑制率明显高于单纯用药各组,差异具有显著性(p<0.05)。结论:4种化疗药物与NS特异性RNAi联合对Eca-109细胞都具有一定的协同抑制作用。 Objective:To study the effect of Nucleostemin (NS) specific RNA interference combined 4 kinds of anticancer drugs on the cell proliferation in human sophageal carcinoma cell lines. Methods: 1. An NS - specific siRNA expression vector was constructed to transfect Eca - 109 cell. 2. The groups of the experiment: silencer group( S group) , normal group( N group) , single -drug group, combined -drug group. S group was the human esophageal carcinoma Eca - 109 cells transfected by PCDNA4/C - NS - silencer. N group was the normal human esophageal carcinoma Eca- 109 cells. Single - drug group was the human esophageal carcinoma Eca- 109 cells affected by anticancer drug only. Combined - drug group was the human esophageal carcinoma Eca - 109 cells affect- ed by NS specific RNA interference combined anticancer drugs. 3. To examine the cell proliferation by cell counting the morphological observation on cells under microscope and examining the inhibiting rate of cell proliferation of differential groups. Results:The proliferation velocity of tumor cells of all single - drug groups was obviously lower, but the inhibiting rate of cell proliferation was obviously higher compared with the cells of all the combined - drug groups, and the difference has significance( p 〈 0.05 ). Conclusion: 1. The association of all the 4 kinds of anticancer drugs with NS specific RNA interference has definite synergistic inhibitory effect.
