摘要
目的进一步了解浙江宁海卫氏并殖吸虫遗传背景。方法对宁海小汀、宁海西溪(冷风洞)和宁海余山卫氏并殖吸虫不同种群进行核糖体DNA第二间区(ITS2)和线粒体细胞色素C氧化酶亚单位(CO1)基因序列分析研究。结果浙江宁海三地卫氏并殖吸虫ITS2序列有363个碱基对,肺型肺吸虫和非肺型肺吸虫ITS2序列完全相同,与泰国种群(AF159604,Nakorn Nayok省)核酸同源性为97.5%,两者差异有显著意义。浙江宁海小汀和宁海余山二地卫氏并殖吸虫CO1序列有390个碱基对,其中3个核酸位点不同(No.49,178,331);浙江宁海小汀种群与中国闽清种群(U97209)和泰国种群(U97212)核酸同源性分别为99.5%和89.0%。结论浙江宁海卫氏并殖吸虫属亚洲东北组群。
Objective To further understand the genetic background of Paragonimus westermani in Ninghai, Zhejiang province. Methods The metacercariaes were collected from Xiaoting, Xixi and Yushan vil- lages in Ninghai County for ITS2 and CO1 gene sequence analysis. Results There were 363 bp of ITS2 sequence of the Paragonimus westermani in the three villages. The ITS2 sequence of two types of P.westermani in Ninghai was equal completely. The significant difference of ITS2 sequence was found between the populadora from Zhejiang and Thailand (AF159604,Nakom Nayok Province). The homology of nucleic acid was 97.5%. There were 390 bp of CO1 sequence in Xiaoting and Yushan village, Ninghai County, and 3 nucleotide codes were different (No.49,178,331). The homology of nucleic acid between Xiaoting village, Ninghai County and Mingqin China (U97209), Thailand (U97212) was 99.5% and 89.0%, respectively. Conclusion The P.westermani in Ninghai, Zhejiang belonged to North-east group of Asia.
