摘要 目的建立大鼠腹部异位心脏移植模型,探索稳定的手术方法。方法实验分A组(对照组)、B组(实验组)和C组(空白组),各30只,A组、B组行心脏移植,C组未行手术。A组采用传统灌注方法;B组采用改进的心脏移植模型灌注方法,采用肝素生理盐水从肝上下腔静脉灌注心脏,同时剪断腹主动脉放血,至心脏完全停跳停止灌注。心脏移植成功后检测外周血超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果B组采用改进方法后,整个灌注过程的平均时间在2 min内,较传统的灌注法缩短10min以上,热缺血时间平均缩短25 s,且SOD活性和MDA含量与A组的差异有显著性意义(P<0.05)。结论改进的灌注方法简单实用,既缩短了灌注时间、热缺血时间和手术时间,又减轻了缺血再灌注损伤,提高了成功率。 Objective To establish the model of abdominal heterotopic heart transplantation and grope steady operation methods. Methods The heart transplantation was performed in the control group (group A) and experiment group (group B) with 30 rats in each group. Another 30 animals not performed the operation were as the blank group (group C) . During the procurement of the donor heart saline solution containing heparitin was injected into the inferior vena cava of liver. At the same time abdominal aorta was transected. Then heart was perfused until it stopped. The activity of superoxide dismutase (SOD) and content of malondialdehyde (MDA) in blood was measured after transplantation. Results The total perfusion time of the graft was shortened within 2 min and hot ischemia timg was shortened within 25 s in the group B. The activity of SOD and content of MDA between two groups was significantly different (P 〈0.05). Conclusion The perfusing method improved with new technique is quicker and more effective than the traditional one and results in the successful rate improvement.
