摘要
通过对GM-CSF工程菌W3100发酵条件的研究,优化了培养及表达条件,使工程菌W3100在30L发酵罐可表达分泌型GM-CSF1.5g/L左右,占菌体总蛋白的25%以上,纯化后的比活大于2.0×107。
The culture and expression coditions of secretory recombinant E. coli W3100/pGM-CSF were optimized during the pilot fermention process. More than 25 % (about 1. 5g/L) of total somatic proteins of secrtory GM-CSF were expressed in W3100 culture in 30L fermenter under the improved conditions. The specific activities of product reached over 2.0×107/ mg protein after being purified,
