摘要
建立了反相离子对色谱法同时测定万乳康样品中淫羊藿苷和盐酸左旋咪唑的含量。本法使用Shim—packVP—ODS色谱柱(250×4.6mm,5μm),该流动相为65%甲醇(V/V),10.0mmol/L的十二烷基磺酸钠,PH=3.0;等度洗脱样品中的淫羊藿苷和盐酸左旋咪唑。紫外检测波长270nm。结果表明:淫羊藿苷测定的线性范围为2.73~54.58μg/mL,加标回收率为99.45%~100.69%,理论塔板数达6641;盐酸左旋咪唑测定的线性范围为26.23~524.54μg/mL,加标回收率为99.58%~100.23%,理论塔板数5564。该方法可用于淫羊藿提取物和万乳康样品中的淫羊藿苷和盐酸左旋咪唑的测定。
A reversed phase ion-pair isocratic elution liquid chromatographic method has been developed and validated for Icariin and Levamisole Hydrochloride. Mixtures of methanol-phosphoric acid aqueous solution containing sodium dodecane sulfonate as the ion-pairing reagent were used as the mobile phase. The icariin, levamisole hydrochloricde have been separated on Shim-pack VP-ODS (250×4.6 mm i. d. , 5 μm) with UV absorbance detector(λ=270 nm). The mobile phase contains 65% methanol(V/V), 10 mmol/L sodium dodecane sulfonate with pH=3.0. The linear range was 2.73-54.58μg/mL for icariin and 26.23-524.54μg/mL for levamisole hydrochloride,the recovery was 99.45%-100.69% for icariin and 99.58%-100.23% for levamisole hydrochloride,theoretical plate number was 6 641 for icariin and 5 564 for levamisole hydrochloride, respectively. The method has been applied to the determination of Herba Epimedii extracts and Wan Ru Kang samples with satisfactory results.
