摘要
利用光谱修正技术研究脱氧核糖核酸(DNA)与5-Br-PAN-S(L)分子间的相互作用,通过pH 8.04的介质中DNA与5-Br-PAN-S反应的光谱研究,测定了结合产物的物理化学参数:10℃时探针L与DNA的结合比为4.3,结合常数K=0.6118×106;30℃时探针L与DNA的结合比为3.7,结合常数K=0.6769×106。此结合反应符合Langmuir吸附方程,该方法可应用于痕量DNA的测定。
The interaction of molecules of DNA with 5-Br-PAN-S was studied by applying the spectral correction technique. Through absorptio-spectrometry, In a buffer medium of pH 8. 04, the physical chemical parameters of the combination product were studied by spectro-photometry. The combination ratio between 5-Br-PAN-S and DNA (L : DNA) at 10 ℃ was found to be 4.3, with combination constant of 0. 611 8×10^6 while at 30 ℃, the ratio was 3. 7 with combination constant of 0. 676 9 ×10^6. It was found that the interaction conformed with the Langmuir adsorption equation. This interaction was feasible to be used for the determination of DNA.
