摘要
为检验尿激酶在临床应用时对小梁细胞有无不良影响。本研究应用组织培养人眼小梁细胞,观察不同浓度尿激酶对细胞形态的影响,并测定其对细胞DNA合成时氚(标胸腺嘧啶核着(3H-TdR)掺入的量,来判断药物对细胞增殖功能的影响。结果示5000u/ml浓度作用6小时细胞即出现胞突回缩、胞体皱缩等变化,至48小时细胞死亡;5000和500u/ml浓度均显著抑制DNA合成。提示在应用尿激酶冲洗前房积血时,一定要冲洗干净残留药物,以免对小梁细胞造成损害。
We evaluated the effect of urokinase on the morphologyDNA synthesis of cultured human trabecular meshwork cells in 3rd generation . Expeare to urokinase at a concerntration of 5000u/ml for 6 hours led to retraction of cytoplasmic procress-es and rounding of cell profile, and exposure for, 48 hours caused call death. Tritiated thymidine incorporation was determined as an index of DNA synthesis, which was significantlyinhibited by 5000 u/ml adn 500u/ml urokinase The auther suggested that the anterior chamber must be washed with BSS to rid of the rest of urokinase when 1000 u/ml urokinase was used for irrigation of hyphema so as to prevent trabecular meshwok cell from damage of the residual drug.
