血小板活化因子拮抗剂ONO-6240对淋巴细胞分泌白细胞介素-5的影响

EFFECT OF A PLATELET ACTIVATING FACTOR ANT AGONIST ONO 6240 ON THE PRODUCTION OF INTERLEUKIN 5 BY LYMPHOCYTES

为研究ＯＮＯ－６２４０对淋巴细胞（ＬＣ）产生白细胞介素－５（ＩＬ－５）的影响，分离培养致敏小鼠ＬＣ，于体外培养时加入不同浓度的ＯＮＯ－６２４０，以ＥＬＩＳＡ法测定ＬＣ培养上清液中ＩＬ－５的含量。结果发现，ＯＮＯ－６２４０能抑制ＬＣ产生ＩＬ－５，并且呈明显的剂量相关性，而这种抑制作用至少可以维持７２ｈ。提示ＯＮＯ－６２４０由于能抑制ＩＬ－５的产生，从而可用于治疗某些过敏性疾病。

In order to investigate the effect of a platelet activating factor antagonist ONO 6240 on the secretion of interleukin(IL) 5 by lymphocytes,lymphocytes isolated from mice spleen were cultured in vitro supplemented with different doses of ONO 6240.ELISA method was used to detect the concentrations of IL 5 in the supernatants of cultured lymphocytes.The result showed that ONO 6240 could inhibit the production of IL 5 by lymphocytes,and such inhibition showed a dose—effect relationship.This inhibition lasted as least 72 h.Our results demonstrated that ONO 6240 may be of some value in treating certain kinds of human allergic diseases because of its ability in inhibiting the productuon of IL 5.
