摘要
目的观察干扰素α-2b对大鼠实验性胰腺纤维化Ⅲ型胶原和纤维连接蛋白合成的影响。方法采用每周2次腹腔内注射二乙基二硫代氨基甲酸盐(diethyldithio carbamate,DDC)1000mg/kg体重诱导健康雄性Wistar大鼠胰腺纤维化。干扰素α-2b治疗组(n=10)在造模日起每天1次皮下注射干扰素α-2b100000U,连续6周。对照组(n=10)注射等容积生理盐水。6周末处死大鼠,取新鲜胰腺组织,10%甲醛溶液固定,常规石蜡包埋切片,观察病理改变。免疫组织化学染色检测Ⅲ型胶原和纤维连接蛋白。结果干扰素α-2b治疗组腺泡细胞炎症反应和纤维化程度较对照组减轻;干扰素α-2b治疗组Ⅲ型胶原和纤维连接蛋白量分别为88.14±42.58和2.37±1.56,较对照组的59.95±25.50和1.50±0.31显著减少(P〈0.05)。结论干扰素α-2b可能通过减少胰腺组织内Ⅲ型胶原和纤维连接蛋白的合成起到治疗胰腺纤维化的作用。
Objective To investigate the effect of interferon α-2b on the expression of collagen Ⅲ and fibronectin in the pancreatic tissue of rats with pancreatic fibrosis. Methods The pancreatic fibrosis model was induced by intra abdominal injection with diethyldithio carbamate solution 1 000 mg/kg weight twice per week. Twenty Wistar rats were randomly divided into 2 groups (n=10) : control group and interferon α-2b treatment group. The rats in α-2b interferon treatment group were hypodermic injected with α-2b interferon 100 000 unit/kg weight once per day for 6 weeks, while the control group was treated with the same volume saline. At the end of the 6th week, these rats were sacrificed and the pancreatic tissues were taken to do the following experiments: 1. HE staining was used to evaluate the degree of inflammation. 2. The expression of collagen Ⅲ and fibronectin were detected by immunohistochemistry. Results Both the degree of inflammation and fibrosis in the control group were more serious than the α-2b interferon treatment group. Moreover the expression of collagen Ⅲ (88.14 ± 42.58) and fibronectin (2. 37 ± 1. 56) in interferona-2b treatment group was markedly reduced as compared with those in the control group (59.95 ± 25.50 and 1. 50 ± 0.31, P 〈 0.05). Conclusions Interferon α-2b may decrease the expression of collagen Ⅲ and fibronectin, which may play a role in antifibrosis therapy of pancreas.
