摘要
目的:观察白藜芦醇(Res)对重症急性胰腺炎(SAP)大鼠肺泡巨噬细胞功能的影响。方法:36只大鼠随机分为假手术组、重症急性胰腺炎模型组(SAP)和白藜芦醇治疗组3组。制模后6h行肺、胰腺组织病程学检查。经支气管肺泡灌洗获取肺泡巨噬细胞,检测支气管肺泡灌洗液中蛋白含量、肺泡巨噬细胞肿瘤坏死因子α(TNF-α)及一氧化氮(NO)水平。以反转录聚合酶链反应(RT-PCR)法测定肺泡巨噬细胞TNF-αmRNA、诱导型一氧化氮合酶(iNOS)mRNA表达情况。结果:Res处理组6h肺支气管肺泡灌洗液中蛋白含量为(844.7±198.2)μg/mL,显著低于SAP组(P<0.05);Res处理组肺泡巨噬细胞分泌TNF-α和NO水平分别为(544.7±98.4)pg/mL和(31.7±9.8)μmol/L,显著低于SAP组(P<0.05)。Res组肺泡巨噬细胞TNF-αmRNA、iNOSmRNA的表达相对强度,亦显著低于SAP组。SAP大鼠各组指标与假手术组相比差异均有显著性意义(P<0.05)。结论:Res能显著减轻SAP时肺和胰腺组织损伤,其机制与抑制巨噬细胞功能,抑制TNF-α和iNOSmRNA表达有关。
To investigate the effect of resveratrol(Res) on function of alveolar macrophage in lung of rats with severe acute pancreatitis-associated lung injury Methods Thirty-six rats were divided into three groups: control group, SAP model group and Res treated group. Rats were sacrificed at 6 h after inducing SAP model. The pathological changes of the lung and pancreas were observed under light microscopy. Alveolar macrophages were harvested by brochoalveolar lavage. The protein content of lavage fluids, the tumor necrosis factor-or (TNF-α) and nitric 0xide(NO) secreted by alveolar macrophages were determined. The expression of TNF-α mRNA and inducible nitric oxide synthase( iN-OS) mRNA was measured with reverse transcription-polymerase chain reaction(RT- PCR)technique. Results The protein content of lavage fluids, TNF-α and NO level in Res-treated group was 844.7 ± 198.2 ptg/ml, 544.7± 98.4 pg/ ml and 31.7 ±9.8 twnoL/L, significantly lower than those in SAP groups( P 〈 0.05). And the expression of TNF-α mR-NA and iNOS mRNA in Res group was significantly lower than that in SAP group( P 〈 0.05). All the parmneters of SAP groups compared to sham operation group were statistically significant ( P 〈 0.05), Conclusion Res can relieve the injury of lung and pancreas in rats with SAP, the mechanism of which may be related to the inhibitory effect of Res on the function of alveolar macrophages in lung by inhibition of expression of TNF-α and iNOS mRNA,
