RNA干扰的实验方法及应用

摘要 RNA干扰(RNA interference,RNAi)就是把与内源mRNA编码区同源的长度为19-23bp的外源双链RNA(double stranded RNA,dsR-NA)导人细胞中,通过一系列细胞内反应引发细胞中相应mRNA的降解,从而导致特定基因表达沉默的一种技术。是真核生物中普遍存在的抵抗病毒人侵,抑制转座子活性及基因表达的监控机制。由于RNAi可以作为一种简单、有效代替基因敲除的遗传工具,在功能基因组学领域掀起一场真正的革命,并彻底改变这个领域的研究步伐。本文从RNAi的发现着手,对其原理、实验方法及应用作了系统介绍。
