免疫组化选择性DNA序列测定技术探讨

Investigation on molecular method of immunohisto selective DNA sequencing

为进一步提高ＤＮＡ序列分析方法的敏感性，用免疫组化选择性ＤＮＡ序列测定技术（ＩＨＳＳ），对食管癌前病变组织中肿瘤抑制基因Ｐ５３免疫阳性细胞进行测定。结果：ＩＨＳＳ法可分离和提高Ｐ５３免疫阳性细胞的数量，并可对发生极少量Ｐ５３蛋白聚集阳性细胞的病变组织进行ＤＮＡ序列测定。这一新方法还可应用于其它分子生物学的研究。

In order to increase the sensitivity of DNA sequencing analysis,an immunohisto selective DNA sequencing method was developed to analyse P53 immunostain positive cells in esophageal precancerous lesions.The results showed that IHSS method can isolate and enrich the P53 immunostain positive cells and to analyse DNA sqeuencing on lesions with few P53 protein accumulation positive cells.This new method may also be a general method for other molecular analysis.
