摘要
应用ras癌基因产物P21蛋白一段氨基酸序列的合成肽的单克隆抗体EB2411和erbB2癌基因产物P185抗体,对人胃癌、癌旁和正常组织中ras和erbB2的表达进行免疫组化研究,结果:P21在正常、癌旁和癌组织的阳性率分别为133%、793%和689%;阳性细胞数分别为(40±15),(219±104)和(1472±400)mm-2,三者之间呈显著性差异(P<001)。P21的表达与肿瘤的分化类型无显著性差异。早期胃癌(病变局限于粘膜层和粘膜下层)P21表达高于进展期胃癌。高分化腺癌中P185IR阳性细胞数明显多于中、低分化癌。Ⅰ、Ⅱ期胃癌P185阳性细胞数较少,而Ⅲ、Ⅳ期显著增多。提示:P21参与胃癌的早期发生,而P185与胃癌的晚期发展有关,并参与胃癌细胞的分化调节。
The expression of ras P21 and erbB 2 P185 in human gastric cancer, adjacent to cancer and normal mucosa were examined by using monoclonal antibody EB2411 to a synthetic peptide reflecting amino acid position 56~64 of ras P21 protein and multiclonal antibody of P185 with immunohistochemical technique.The results showed that the expression rates of P21 in the normal gastric mucosa, adjacent to cancer and gastric cancer were 13.3%, 79.3% and 68.9% respectively. The positive cells in gastric cancer, adjacent to cancer and normal mucosa were (147.22± 39.96) mm -2 ,(21.89±10.38)mm -2 and (3.96±1.52) mm -2 respectively. There was significant difference among the three kinds of tissues. However, there was no obvious relation between the expression of P21 and the differentiation types of gastric cancer ( P <0.05), and it demonstrated a heterogeneous expression in gastric cancer. The expression of P185 erbB 2 was similar to that of P21 ras in gastric cancer, adjacent to cancer and normal ti ssue. But it was significantly enhanced in well differentiated gastric cancer as compared with moderately and poorly differentiated gastric cancers. The expression of P185 was increased in clinic stages Ⅲ and Ⅳ more than that in stages Ⅰ and Ⅱ. The results indicated that both the expression of P21 and P185 were enhanced in cancerous tissues. There was an increased expression of P185 in advanced and well differentiated gastric cancer, while P21 enhanced the expression in early gastric cancers, and there was no obvious correlation with the differentiated types of gastric cancers.
