摘要
目的:探讨survivin在三苯氧胺(TAM)诱导乳腺癌细胞凋亡及其细胞耐药中的作用。方法:用RT-PCR和Western-blot检测survivin mRNA和蛋白表达,四唑蓝(MTT)比色法测定细胞增殖活性,用流式细胞仪检测细胞凋亡。结果:TAM浓度在1μg/ml时,发挥最大的促凋亡和抑增殖活性,且sur-vivin mRNA及蛋白表达下降最明显,细胞生长抑制率随着TAM浓度的增加和作用时间的延长而增加。结论:TAM诱导乳腺癌细胞凋亡与survivin有关,survivin与肿瘤细胞的凋亡和增殖有关,并参与肿瘤细胞耐药,survivin可能是乳腺癌基因治疗的一个靶点。
Objective:To study tamoxifen(TAM) inducing growth arrest and apoptosis and explore chemoresistance with survivin in breast cancer cell line.Method:Detect apoptosis by flow cytometry and measure cell proliferation activity by MTT assary in breast cancer. The cultured MCF-7 cells was treated with TAM.Then,the expression of survivin mRNA and protein by MCF-7 cells was evaluated by semi-quantitative reverse-transcription polymerase chain reaction (RT-PCR) and western blot analysis.Results:When tamoxifen was 1μmol,promoting apoptosis,inhibiting proliferation activity and survivin mRNA protein expression singnificantly decreasring was detected in the human breast cancer cell line.Conclusion:Apoptosis induced by tamoxifen in breast cancer was associated with survivin that plays important role in regulation of cell cycle.survivin was related to the apoptosis and proliferation of carcinoma cells and drug resistance.Apoptosis induced by tamoxifen in breast cancer has been associated with survivin.survivin could be a target in gene therapy.
