毛冬青甲素对小鼠血浆及兔血小板内环磷酸腺苷水平的影响 被引量:1

摘要 毛冬青甲素对冠心病等疾病有明显的疗效,且对血小板的凝集有抑制作用,为了进一步探讨其抑制作用的机理。本实验从毛冬青甲素对血浆 cAMP 及血小板内 cAMP 含量的变化进行了研究。实验结果表明,血浆 cAMP 的含量给药组与对照组比较,差异无统计学意义(P>0.05)。而血小板内 cAMP 的含量是给药组高于对照组(P<0.025)。提示毛冬青甲素对血小板的抑制作用是通过增加血小板内 cAMP的含量来实现的。说明其药理作用机理之一与增加血小板内 cAMP 水平有关。这一研究结果有可能为用活血化瘀法则防治冠心病等疾病提供分子药理学的依据。 To investigate the mechanism of Ilexonin A(IA)in increasing the cAMPlevel of platelets,the activity of phosphodiesterase(PDE)was calculated bytesting the residual of cAMP after its hydrolysis by PDE,and the effectof IA in different doses on the activity of PDE was also studied.The resultsshowed that the amount of cAMP hydrolyzed in IA solutions of 0.1g/L and0.5g/L were 91.90±7.91 pmol and 66.40±2.84 pmol respectively,P<0.05 andP<0.01 as compared with the control group(128.00±7.38 pmol).It isindicated that IA can inhibit the activity of PDE and it increases the cAMPlevel in platelet and inhibits the aggregation of platelet through the inhi-bition of the activity of 3′—5′ cyclic nucleotide PDE.
