摘要
将多聚甲醛固定的裸甲藻(Takayama pulchellum,TPXM)细胞采用离心法收集,进行小梯度的酒精结合离心法逐级脱水,临界点干燥并真空喷金后,能够在扫描电镜(SEM)下很好地显示出T.pulchellum细胞的完整形态和细微结构,如纵鞭毛、横沟、顶沟、顶突、带状物、藻体线纹、腰鞭毛等;同时环境扫描电镜(ESEM)也能对裸甲藻进行较好的形态学观察,但环境样品的ESEM和SEM电镜观察条件尚需要进一步完善.
Takayama pulchellum cells fixed by paraformaldehyde (1% final concentration) were collected by centrifugal methods, dehydrated by small grads ethanol concentration and centrifugated again, then evaporation under critical point, spray gold in vacuum. The integrated shown clearly under scanning electron microscope, and detailed structure characteristics of T. pulchellum cells were such as longitudinal flagella, transverse groove, apical groove, apical carina, cingulum, spiral marking, transverse flagella etc. In the meantime, environmental scanning electron microscope was a good tool to observe the morphological characteristics of T. pulchellum cells, but the protocols of ESEM and SEM used to investigate natural samples still need to be optimized.
